On first boot you will be prompted to configure a controller in EmulationStation. Once you finish configuring your controller, multiple configuration profiles will be automagically generated for most of the systems in RetroPie. How does one define a game controller? It could simply be defined as an input device for a machine on which you play a game. No, a game controller is more specific than that.
