To determine whether extended times of incubation could increase the percentage of non-motile bacteria in MC1000 biofilm, we tried to recover isolates from 1-month old biofilms. In duplicate experiments, the biofilm disintegrated after 2weeks. At 4weeks, we recovered only between 10 4 and 10 5 colony forming units (CFU) from a surface area of 28.3cm 2 (data not shown).

J Bacteriol 193, 1600–1611 (2011). The first set of experiments led to the conclusion that the biofilm environment may favor a mixture of motile and non-motile bacteria. From biofilms formed by the highly motile MC1000, we recovered 62 non-motile isolates, but no hyper-motile ones. We conclude that moderate selective pressure towards the opposing motility phenotype might operate in this biofilm. If this were true, then the biofilm environment may favor a mixture of motile and non-motile bacteria. To test this hypothesis, we initiated biofilms with MC1000 or with a mixture of MC1000 and the non-motile BP19 and followed the composition of the biofilm for about one month. The biofilm formed by the former degraded, whereas the latter formed a biofilm that maintained its integrity for a month with a mixture of both strains for most of the 1 month time period of the experiment (Fig. 1).

In 1971, the Harley-Davidson FX ″Super Glide″ was introduced as a ″factory chopper″, combining the FL frame and drive train with the smaller XL front end, basically creating the third, intermediate model range FX that sold and sells well. Since then, the FL prefix indicates that the traditional 16-inch front wheel and big fenders are used on ″Dresser″ Touring series or the traditional styling ″Heritage″ variants of the Softail series, while FX is used on sportier models or the chopper-like Softails with rather thin front wheels. In contrast, the loss of fliZ resulted in a consistent reduction in motility, except for the flhC EC strain. However, as the flhC EC strain was already impaired in motility, it is possible that the resolution of the motility assay was unable to identify differences in the ∆ fliZ mutant. Flagellar gene expression activity did, however, suggest a 2-fold drop in P fliC expression in the flhC EC ∆ fliZ strain as compared to the otherwise wild-type (Fig. 3B).
